[Chemical Knowledge]:Dry blending and wet blending: two common ways of pharmaceutical granulation

In the process of drug manufacturing, granulation is a very important link. It is the process of converting raw material powder into granular solid preparations through processing methods. In the granulation process, dry mixing and wet mixing are two common ways. Although they are all granulation methods, there are obvious differences in the operation process, applicable raw materials and final product characteristics.

First, let's look at dry mix granulation. Dry mixing granulation refers to mixing powder raw materials with auxiliary materials under anhydrous or low water content conditions, and then processing the preparation through a tablet press or granulator. The advantages of dry mixing granulation are simple operation, low cost and wide application range, so it is widely used in the pharmaceutical industry. It is suitable for handling water-sensitive materials, but also for some of the more difficult to process raw materials, such as some easy adhesion, easy to form agglomerates of powder raw materials. However, dry mixing granulation also has some limitations, such as in the case of some small particles, easy to break, will lead to poor uniformity of the particles, and the edge of the particles easy to produce wear phenomenon.

followed by wet mix granulation. Wet mixing granulation refers to mixing the powder raw materials with the auxiliary materials under the condition of adding an appropriate amount of water or solvent, and then processing the preparation by wet ball milling or wet granulator. Compared with dry mixing granulation, the quality of the particles in wet mixing granulation is more uniform, the surface of the particles is smoother, and the compactness and mechanical strength of the particles are better. Therefore, wet mixing granulation is a better choice in some occasions with high quality requirements for granules, such as the production of slow-release granules and controlled-release granules. However, wet mixing granulation also has some disadvantages, such as the need for additional drying steps, the operation is relatively complicated, and the range of raw materials that can be applied is relatively narrow.

In summary, dry blending and wet blending are two common methods in pharmaceutical granulation. Dry mixing granulation has a wide range of applications, low cost, simple operation, and is suitable for handling water-sensitive materials. Wet mixing granulation has more advantages in particle quality and mechanical strength, and is suitable for occasions with high requirements on particle quality. In actual production, pharmaceutical companies need to choose the appropriate granulation method according to their own product characteristics and production process to ensure the quality and stability of the final product.
